NM_013372.7(GREM1):c.506G>A (p.Arg169Lys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R169K variant (also known as c.506G>A), located in coding exon 1 of the GREM1 gene, results from a G to A substitution at nucleotide position 506. The arginine at codon 169 is replaced by lysine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
