Uncertain significance — the classification assigned by Ambry Genetics to NM_012108.4(STAP1):c.157A>C (p.Thr53Pro), citing Ambry Variant Classification Scheme 2023: The p.T53P variant (also known as c.157A>C), located in coding exon 2 of the STAP1 gene, results from an A to C substitution at nucleotide position 157. The threonine at codon 53 is replaced by proline, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
