NM_007294.4(BRCA1):c.4365A>C (p.Leu1455Phe) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L1455F variant (also known as c.4365A>C), located in coding exon 12 of the BRCA1 gene, results from an A to C substitution at nucleotide position 4365. The leucine at codon 1455 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
