NM_007194.4(CHEK2):c.1047G>C (p.Lys349Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K349N variant (also known as c.1047G>C), located in coding exon 9 of the CHEK2 gene, results from a G to C substitution at nucleotide position 1047. The lysine at codon 349 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
