Uncertain significance — the classification assigned by Ambry Genetics to NM_006904.7(PRKDC):c.2732T>C (p.Leu911Pro), citing Ambry Variant Classification Scheme 2023: The p.L911P variant (also known as c.2732T>C), located in coding exon 24 of the PRKDC gene, results from a T to C substitution at nucleotide position 2732. The leucine at codon 911 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
