Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_006432.5(NPC2):c.223C>T (p.His75Tyr), citing Ambry Variant Classification Scheme 2023: The p.H75Y variant (also known as c.223C>T), located in coding exon 3 of the NPC2 gene, results from a C to T substitution at nucleotide position 223. The histidine at codon 75 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
