NM_006265.3(RAD21):c.690T>A (p.Asp230Glu) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D230E variant (also known as c.690T>A), located in coding exon 6 of the RAD21 gene, results from a T to A substitution at nucleotide position 690. The aspartic acid at codon 230 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
