Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_006258.4(PRKG1):c.119A>T (p.Gln40Leu), citing Ambry Variant Classification Scheme 2023: The p.Q40L variant (also known as c.119A>T), located in coding exon 1 of the PRKG1 gene, results from an A to T substitution at nucleotide position 119. The glutamine at codon 40 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
