Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.5177G>A (p.Cys1726Tyr), citing Ambry Variant Classification Scheme 2023: The p.C1726Y variant (also known as c.5177G>A), located in coding exon 39 of the POLE gene, results from a G to A substitution at nucleotide position 5177. The cysteine at codon 1726 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
