Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.3944T>C (p.Leu1315Pro), citing Ambry Variant Classification Scheme 2023: The p.L1315P variant (also known as c.3944T>C), located in coding exon 31 of the POLE gene, results from a T to C substitution at nucleotide position 3944. The leucine at codon 1315 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
