NM_006073.4(TRDN):c.14C>T (p.Thr5Ile) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T5I variant (also known as c.14C>T), located in coding exon 1 of the TRDN gene, results from a C to T substitution at nucleotide position 14. The threonine at codon 5 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
