Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005751.5(AKAP9):c.7124A>G (p.His2375Arg), citing Ambry Variant Classification Scheme 2023: The p.H2375R variant (also known as c.7124A>G), located in coding exon 31 of the AKAP9 gene, results from an A to G substitution at nucleotide position 7124. The histidine at codon 2375 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
