Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.379G>T (p.Val127Phe), citing Ambry Variant Classification Scheme 2023: The p.V127F variant (also known as c.379G>T), located in coding exon 4 of the RAD50 gene, results from a G to T substitution at nucleotide position 379. The valine at codon 127 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
