NM_000057.4(BLM):c.77G>A (p.Ser26Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S26N variant (also known as c.77G>A), located in coding exon 1 of the BLM gene, results from a G to A substitution at nucleotide position 77. The serine at codon 26 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
