Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_005120.3(MED12):c.6334C>G (p.Gln2112Glu), citing Ambry Variant Classification Scheme 2023: The p.Q2112E variant (also known as c.6334C>G), located in coding exon 43 of the MED12 gene, results from a C to G substitution at nucleotide position 6334. The glutamine at codon 2112 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
