NM_004655.4(AXIN2):c.556G>A (p.Ala186Thr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A186T variant (also known as c.556G>A), located in coding exon 1 of the AXIN2 gene, results from a G to A substitution at nucleotide position 556. The alanine at codon 186 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
