NM_004415.4(DSP):c.8134G>A (p.Ala2712Thr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A2712T variant (also known as c.8134G>A), located in coding exon 24 of the DSP gene, results from a G to A substitution at nucleotide position 8134. The alanine at codon 2712 is replaced by threonine, an amino acid with similar properties. This alteration has been reported in a dilated cardiomyopathy (DCM) cohort (Elliott P et al. Circ Cardiovasc Genet, 2010 Aug;3:314-22). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
