Uncertain significance — the classification assigned by Ambry Genetics to NM_004336.5(BUB1):c.3088G>C (p.Glu1030Gln), citing Ambry Variant Classification Scheme 2023: The p.E1030Q variant (also known as c.3088G>C), located in coding exon 25 of the BUB1 gene, results from a G to C substitution at nucleotide position 3088. The glutamic acid at codon 1030 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
