Uncertain significance — the classification assigned by Ambry Genetics to NM_004336.5(BUB1):c.1532C>G (p.Ser511Cys), citing Ambry Variant Classification Scheme 2023: The p.S511C variant (also known as c.1532C>G), located in coding exon 14 of the BUB1 gene, results from a C to G substitution at nucleotide position 1532. The serine at codon 511 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
