NM_004336.5(BUB1):c.1046T>A (p.Val349Glu) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V349E variant (also known as c.1046T>A), located in coding exon 10 of the BUB1 gene, results from a T to A substitution at nucleotide position 1046. The valine at codon 349 is replaced by glutamic acid, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
