Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.4349G>C (p.Gly1450Ala), citing Ambry Variant Classification Scheme 2023: The p.G1450A variant (also known as c.4349G>C), located in coding exon 29 of the ALK gene, results from a G to C substitution at nucleotide position 4349. The glycine at codon 1450 is replaced by alanine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
