Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.4302G>T (p.Glu1434Asp), citing Ambry Variant Classification Scheme 2023: The p.E1434D variant (also known as c.4302G>T), located in coding exon 29 of the ALK gene, results from a G to T substitution at nucleotide position 4302. The glutamic acid at codon 1434 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
