Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.3705T>G (p.Cys1235Trp), citing Ambry Variant Classification Scheme 2023: The p.C1235W variant (also known as c.3705T>G), located in coding exon 24 of the ALK gene, results from a T to G substitution at nucleotide position 3705. The cysteine at codon 1235 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
