NM_004304.5(ALK):c.2158C>G (p.Leu720Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L720V variant (also known as c.2158C>G), located in coding exon 12 of the ALK gene, results from a C to G substitution at nucleotide position 2158. The leucine at codon 720 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
