Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.1031A>C (p.His344Pro), citing Ambry Variant Classification Scheme 2023: The p.H344P variant (also known as c.1031A>C), located in coding exon 4 of the ALK gene, results from an A to C substitution at nucleotide position 1031. The histidine at codon 344 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
