Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003977.4(AIP):c.720C>G (p.Cys240Trp), citing Ambry Variant Classification Scheme 2023: The p.C240W variant (also known as c.720C>G), located in coding exon 5 of the AIP gene, results from a C to G substitution at nucleotide position 720. The cysteine at codon 240 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
