Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_003611.3(OFD1):c.2742A>T (p.Lys914Asn), citing Ambry Variant Classification Scheme 2023: The p.K914N variant (also known as c.2742A>T), located in coding exon 20 of the OFD1 gene, results from an A to T substitution at nucleotide position 2742. The lysine at codon 914 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
