Uncertain significance — the classification assigned by Ambry Genetics to NM_003579.4(RAD54L):c.872G>A (p.Gly291Asp), citing Ambry Variant Classification Scheme 2023: The p.G291D variant (also known as c.872G>A), located in coding exon 8 of the RAD54L gene, results from a G to A substitution at nucleotide position 872. The glycine at codon 291 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
