Uncertain significance — the classification assigned by Ambry Genetics to NM_003579.4(RAD54L):c.36G>C (p.Lys12Asn), citing Ambry Variant Classification Scheme 2023: The p.K12N variant (also known as c.36G>C), located in coding exon 2 of the RAD54L gene, results from a G to C substitution at nucleotide position 36. The lysine at codon 12 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
