NM_003579.4(RAD54L):c.1027A>G (p.Asn343Asp) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N343D variant (also known as c.1027A>G), located in coding exon 9 of the RAD54L gene, results from an A to G substitution at nucleotide position 1027. The asparagine at codon 343 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
