NM_003476.5(CSRP3):c.180G>A (p.Val60=) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.180G>A variant (also known as p.V60V), located in coding exon 2 of the CSRP3 gene, results from a G to A substitution at nucleotide position 180. This nucleotide substitution does not change the valine at codon 60. This nucleotide position is poorly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will result in the creation or strengthening of a novel splice donor site. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
