NM_001032283.3(TMPO):c.565+1406T>G was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I329M variant (also known as c.987T>G), located in coding exon 4 of the TMPO gene, results from a T to G substitution at nucleotide position 987. The isoleucine at codon 329 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
