Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003239.5(TGFB3):c.971T>A (p.Phe324Tyr), citing Ambry Variant Classification Scheme 2023: The p.F324Y variant (also known as c.971T>A), located in coding exon 6 of the TGFB3 gene, results from a T to A substitution at nucleotide position 971. The phenylalanine at codon 324 is replaced by tyrosine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
