Uncertain significance — the classification assigned by Ambry Genetics to NM_002907.4(RECQL):c.1892T>A (p.Leu631His), citing Ambry Variant Classification Scheme 2023: The p.L631H variant (also known as c.1892T>A), located in coding exon 14 of the RECQL gene, results from a T to A substitution at nucleotide position 1892. The leucine at codon 631 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
