Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_002890.3(RASA1):c.1014G>C (p.Glu338Asp), citing Ambry Variant Classification Scheme 2023: The p.E338D variant (also known as c.1014G>C), located in coding exon 5 of the RASA1 gene, results from a G to C substitution at nucleotide position 1014. The glutamic acid at codon 338 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
