NM_002875.5(RAD51):c.344G>A (p.Gly115Asp) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G115D variant (also known as c.344G>A) is located in coding exon 4 of the RAD51 gene. The glycine at codon 115 is replaced by aspartic acid, an amino acid with similar properties. This change occurs in the first base pair of coding exon 4. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
