NM_002691.4(POLD1):c.1007A>G (p.Gln336Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q336R variant (also known as c.1007A>G), located in coding exon 8 of the POLD1 gene, results from an A to G substitution at nucleotide position 1007. The glutamine at codon 336 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
