NM_002519.3(NPAT):c.3113A>G (p.Lys1038Arg) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K1038R variant (also known as c.3113A>G), located in coding exon 17 of the NPAT gene, results from an A to G substitution at nucleotide position 3113. The lysine at codon 1038 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
