Uncertain significance — the classification assigned by Ambry Genetics to NM_002519.3(NPAT):c.2743G>C (p.Val915Leu), citing Ambry Variant Classification Scheme 2023: The p.V915L variant (also known as c.2743G>C), located in coding exon 13 of the NPAT gene, results from a G to C substitution at nucleotide position 2743. The valine at codon 915 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
