Uncertain significance — the classification assigned by Ambry Genetics to NM_002519.3(NPAT):c.1058A>G (p.Glu353Gly), citing Ambry Variant Classification Scheme 2023: The p.E353G variant (also known as c.1058A>G), located in coding exon 12 of the NPAT gene, results from an A to G substitution at nucleotide position 1058. The glutamic acid at codon 353 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
