Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002354.3(EPCAM):c.736C>G (p.Gln246Glu), citing Ambry Variant Classification Scheme 2023: The p.Q246E variant (also known as c.736C>G), located in coding exon 7 of the EPCAM gene, results from a C to G substitution at nucleotide position 736. The glutamine at codon 246 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
