NM_002354.3(EPCAM):c.160C>G (p.Gln54Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q54E variant (also known as c.160C>G), located in coding exon 2 of the EPCAM gene, results from a C to G substitution at nucleotide position 160. The glutamine at codon 54 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
