This variant lies in the LAMA4 gene (transcript NM_001105206.3) at the canonical splice acceptor site of the intron immediately before coding-DNA position 2814, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.2793-1G>A intronic variant results from a G to A substitution one nucleotide upstream from coding exon 21 of the LAMA4 gene. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice acceptor site and will result in the creation or strengthening of a novel splice acceptor site. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay.
