Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.5807T>C (p.Leu1936Ser), citing Ambry Variant Classification Scheme 2023: The p.L1936S variant (also known as c.5807T>C), located in coding exon 38 of the ATM gene, results from a T to C substitution at nucleotide position 5807. The leucine at codon 1936 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
