Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.7903T>C (p.Tyr2635His), citing Ambry Variant Classification Scheme 2023: The p.Y2635H variant (also known as c.7903T>C), located in coding exon 62 of the FBN2 gene, results from a T to C substitution at nucleotide position 7903. The tyrosine at codon 2635 is replaced by histidine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
