NM_001927.4(DES):c.1363del (p.Asp455fs) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.1363delG variant, located in coding exon 8 of the DES gene, results from a deletion of one nucleotide at nucleotide position 1363, causing a translational frameshift with a predicted alternate stop codon (p.D455Mfs*36). This alteration occurs at the 3' terminus of theDES gene, is not expected to trigger nonsense-mediated mRNAdecay, and only impacts the last 3% of the protein. The exact functional effect of this alteration is unknown. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
