NM_001903.5(CTNNA1):c.591A>T (p.Glu197Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E197D variant (also known as c.591A>T), located in coding exon 5 of the CTNNA1 gene, results from an A to T substitution at nucleotide position 591. The glutamic acid at codon 197 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
