Uncertain significance for Hereditary pancreatitis — the classification assigned by Ambry Genetics to NM_001868.4(CPA1):c.431G>T (p.Ser144Ile), citing Ambry Variant Classification Scheme 2023: The p.S144I variant (also known as c.431G>T), located in coding exon 4 of the CPA1 gene, results from a G to T substitution at nucleotide position 431. The serine at codon 144 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
