NM_000051.4(ATM):c.5174A>G (p.Asp1725Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1725G variant (also known as c.5174A>G), located in coding exon 33 of the ATM gene, results from an A to G substitution at nucleotide position 5174. The aspartic acid at codon 1725 is replaced by glycine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
