NM_001430.5(EPAS1):c.16G>A (p.Glu6Lys) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E6K variant (also known as c.16G>A), located in coding exon 1 of the EPAS1 gene, results from a G to A substitution at nucleotide position 16. The glutamic acid at codon 6 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
